I'm not familiar with game jams, and I've never done any with themes or challenges. I didn't see anything in the rules that specifically state they are required, but I just wanted to ask to be sure.

The theme is required, but isn’t meant to be literal, so some extrapolation is expected. The challenge is optional to some degree, but should at least influence your design.
