While the mission is active and you don't have them, the option should reappear. Remember to collect any other missing pairs too. If it still doesn't work, load an earlier save, but repeating these steps should normally solve it.

For now, wanted pirates can't join your crew; you can only unlock interactions with them. Accept a wanted poster from the Bounty Board in Shells Town, then use RAID from the Captain's Cabin to find the pirate's ship. Damage it without sinking it, board it, and defeat the captain to unlock those interactions.

There isn't a separate unlock for getting every girl's Love and Lust above 50, although some individual interactions have higher requirements.
For now, there isn't a fixed time: the empty-room event is random and has a 2-in-5 chance each time you check. If Robin is there, leave and try again; in the future, I plan to add a schedule showing when she's in her room so this is more intuitive.
