Now that there is the whole simulation part for the NPCs, if I date an NPC via a poly relationship will it be able to date other NPCs and/or invite them to join as a polycule, or they just keep dating the PC and don't see anyone else?

You can form polycule dates, and even tip the odds of them getting to know each other on good terms. That being said, it doesn't seem that they will evolve their relationship without your direct action. If they themselves are poly or open, they may develop other relationships without you getting involved.

Is there any configuration to stop the NPCs from developing other relationships if they are poly or open and already have a relationship with the PC?

Not directly, since it's not counted as 'cheating'. You can try and remove that trait with cheats enabled.
